Artificial sweeteners are listed as ingredients of oral nicotine pouches (ONPs), a new product category with rapidly growing market share. The exact sweetener contents of ONPs remain unknown. Artificial sweeteners in ONPs may facilitate initiation and encourage consumption behavior.
Aims and Methods:
Artificial sweetener contents in major US-marketed ONP brands (Zyn, on!, Velo) were determined by Liquid Chromatography-Mass Spectrometry (LC-MS). Sweetener effects during the initiation of ONP consumption were modeled in single- and two-bottle tests, offering mice ONP extracts calibrated to contain nicotine levels similar to saliva of people who use smokeless tobacco. To examine the contribution of sweet taste perception, consumption behavior was compared between wild-type mice and mice deficient in the sweet taste receptor (Tas1r2-/-).
Results:
Acesulfame-K was detected in on!, Zyn and Velo ONPs (~0.3-0.9 mg/pouch), including products marketed as “Unflavored” or “Flavor ban approved”. In Velo ONPs, sweetened with sucralose (0.6-1.2 mg/pouch), higher nicotine strength products contained higher sucralose levels. Tas1r2-/- mice consumed less ONP extracts than wild-type mice in both sexes. ONP extracts with both higher nicotine and sweetener strengths were tolerated by wildtype mice, but produced stronger aversion in Tas1r2-/- mice.
Conclusions:
ONPs contain significant amounts of artificial sweeteners, with some brands adding more sweetener to ONPs with higher nicotine strengths. Artificial sweeteners, at levels present in ONPs, increase nicotine consumption. Increasing sweetener contents facilitates consumption of ONPs with higher nicotine strengths. Sweetness is a key determinant of ONP use initiation, likely reducing the aversive sensory effects of nicotine and other ONP constituents.
Implications:
Artificial sweeteners such as acesulfame-K or sucralose reduce aversion and facilitate initiation and continued consumption of ONPs. The marketing of some artificially sweetened ONPs as
“Unflavored” of “Flavor ban-approved” suggests that the tobacco industry rejects sweet taste as a determinant for the presence of a characterizing flavor. Sweetness as imparted by artificial
sweeteners in tobacco products needs to be addressed by regulators as a component of a characterizing flavor, with the aim to reduce product appeal and initiation by never users, and

To review the available evidence evaluating the chemicals in refill solutions, cartridges, aerosols and environmental emissions of electronic cigarettes (e-cigarettes).
Methods
Systematic literature searches were conducted to identify research related to e-cigarettes and chemistry using 5 reference databases and 11 search terms. The search date range was January 2
007 to September 2013. The search yielded 36 articles, of which 29 were deemed relevant for analysis.
Results
The levels of nicotine, tobacco-specific nitrosamines (TSNAs), aldehydes, metals, volatile organic compounds (VOCs), flavours, solvent carriers and tobacco alkaloids in e-cigarette refill solutions, cartridges, aerosols and environmental emissions vary considerably. The delivery of nicotine and the release of TSNAs, aldehydes and metals are not consistent across products. Furthermore, the nicotine level listed on the labels of e-cigarette cartridges and refill solutions is often significantly different from measured values. Phenolic compounds, polycyclic aromatic hydrocarbons and drugs have also been reported in e-cigarette refill solutions, cartridges and aerosols. Varying results in particle size distributions of particular matter emissions from e-cigarettes across studies have been observed. Methods applied for the generation and chemical analyses of aerosols differ across studies. Performance characteristics of e-cigarette devices also vary across and within brands.
Conclusions Additional studies based on knowledge of e-cigarette user behaviours and scientifically validated aerosol generation and chemical analysis methods would be helpful in generating reliable measures of chemical quantities. This would allow comparisons of e-cigarette aerosol and traditional smoke constituent levels and would inform an evaluation of the toxicity potential of e-cigarettes

Overall, 11% of the general population have "ever tried" chewing tobacco, snuff, or other smokeless tobacco. Of these, 5% were former users and 3% used smokeless tobacco almost daily in the past year. Rates of its use differed significantly by sex, age group, race, region, and metropolitan area size. Although females were far less likely to try it, those who did were as likely as males to be daily users. Smokeless tobacco users were also more likely to use alcohol, cigarettes, and marijuana. In general, those who used smokeless tobacco almost daily were more likely to report poor health and hospitalization for illness or injury in the past year, even when other substance use was controlled. Smokeless tobacco users also were more likely to report symptoms of depression. Finally, some substituted smokeless tobacco for cigarettes, but youths (12-17 yr old) were more likely than older tobacco users to use both forms of tobacco regularly. This carries significant implications for public health, particularly because a substantial proportion of young individuals who consume white snus had no prior exposure to other tobacco or nicotine products. Consequently, white snus has the potential to attract and introduce new young users to nicotine consumption. This qualitative study aimed to investigate the socio-environmental factors contributing to the consumption of white snus among female adolescents aged between 15 to 18. The data consists of 10 interviews. Guided by the Reciprocal Determinism Model, the study employed thematic analysis to explore the reasons for snus initiation, the continued use of white snus, and the circumstances that could lead to quitting its consumption. Three main themes emerged from the analysis: (1) desirability, (2) exploration and experimentation, and (3) hindrances. The findings elaborated on the socio-environmental factors behind white snus consumption. The marketing and advertisement strategies used by manufacturers were found to reinforce the continued use of white snus among female adolescents. Additionally, economic barriers were identified as the primary motivator for quitting consumption. Peer influence was consistently identified as the trigger for snus initiation. The study also highlighted the ineffectiveness of current regulations in Sweden, as they were easily bypassed, indicating a need for stronger measures to restrict access and limit advertising exposure. As white snus is expanding to countries outside of Sweden, the insights gained from this study can inform the development of early interventions and preventive measures by governments to reduce the likelihood of adolescent nicotine use. Emphasized is the use of time-to-event data on the ages of occurrence of six events in these onset processes. Concepts and methods for the statistical analysis of time-to-event data are demonstrated. The occurrence of events in the smoking onset process are strongly related to increases in the subsequent onset rate for smokeless tobacco use. Compared with before initial smoking has occurred, the onset rates for weekly smokeless tobacco use after initial smoking has occurred are 2.03 (P less than .001) and 6.72 (P less than .001) times as large for males and females, respectively. Furthermore, both initial and weekly use of cigarettes contributes to the risk of subsequent weekly smokeless tobacco use. Conversely, the steps in the onset process of smokeless tobacco use are strongly related to increases in the subsequent smoking onset rate. A three-step approach was taken to analyse the biological effects of NPs and snus extracts in vitro. ToxTracker was used to screen for biomarkers for oxidative stress, cell stress, protein damage and DNA damage. Cytotoxicity, mutagenicity, and genotoxicity were assessed in the following respective assays: Neutral Red Uptake (NRU), Ames and Mouse Lymphoma Assay (MLA). Targeted analysis of phosphorylation signalling and inflammatory markers under non-toxic conditions was used to investigate any potential signalling pathways or inflammatory response. A reference snus (CRP1.1) and four NPs with various flavours and nicotine strengths were assessed. Test article extracts was generated by incubating one pouch in 20 mL of media (specific to each assay) with the inclusion of the pouch material. NP extracts did not induce any cytotoxicity or mutagenic response, genotoxic response was minimal and limited signalling or inflammatory markers were induced. In contrast, CRP1.1 induced a positive response in four toxicological endpoints in the absence of S9: Srxn1 (oxidative stress), Btg2 (cell stress), Ddit3 (protein damage) and Rtkn (DNA damage), and three endpoints in presence of S9: Srxn1, Ddit3 and Rtkn. CRP1.1 was genotoxic when assessed in MLA and activated signalling pathways involved in proliferation and cellular stress and specifically induced phosphorylation of c-JUN, CREB1, p53, p38 MAPK and to a lesser extent AKT1S1, GSK3α/β, ERK1/2 and RSK1 in a dose-dependent manner. CRP 1.1 extracts resulted in the release of several inflammatory mediators including cytokines IL-1α, IL5, IL6, IL8, IL-1RA, MIF and TNF-β, receptor IL-2RA, and growth factors FGF-basic, VEGF and M-CSF. In conclusion these assays contribute to the weight of evidence assessment of the potential comparative health risks of NPs and snus. They originated in Scandinavia, and their distribution is rapidly expanding to other countries, with major tobacco manufacturers selling their products worldwide. Some noteworthy brands include Lyft by British American Tobacco, Velo by RJ Reynolds Vapor Company, Zyn by Swedish Match, Dryft by Kretek International and Nordic Spirits by Japan Tobacco International. Companies promote nicotine pouches as a harmless, tobacco-free alternative and use terms such as “tobacco-free” or “leaf-free” in product marketing. They come in a variety of flavors and are packaged in fancy cans with colorful logos that have great aesthetic appeal. To date, there is a lack of independent research on many aspects of these products, including product ingredients, biomarkers of harm, and evaluation of nicotine delivery.
